Animal experimentation in studying addiction to the newer synthetic analgesics.

چکیده
HE term addiction, when applied to the syndrome resulting from T the prolonged use of morphine and its congeners in man, by coinmon usage involves three principal components, psychic dependence, physical dependence, and acquired toleranee. In view of the fact that the newer synthetic analgesics produce pharmacological effects, many of which are qualitatively similar to those of morphine and its derivatives, it is assumed, for purposes of this discussion, that morphine should stand as the drug of comparison, and that information regarding these three not easily dissociable qualities which constitute addiction to morphine must be obtained for each new agent of this type before it may be safely used clinically. It is clear that clinical facilities for evaluating addiction liability are inadequate to keep pace with the rapid development of the newer synthetic analgesics. This being the case, and recognizing that the final evaluation of addiction liability of any compound proposed for clinical use must be determined in the human species, only that evidence which relates to the following question will be presented and discussed here: Does any infrahuman species exist which parallels normal man in its ability to develop “addiction“ when chronically poisoned with morphine, its congeners, and the newer synthetic analgesics, and, if so, is this parallelism sufficiently striking that this species could be used as a reliable test object for the preliminary scrcening of new compounds? Since comparatively little work has been done in this field with the newer analgesic agents, a large share of the following discussion must center about morphine.

There are currently effective, U.S. Food and Drug Administration (FDA)-approved therapies for alcohol, nicotine, and opioid addiction. In some cases these therapeutics were rationally designed and tested using a combination of various animal models of addiction. In many cases, however, effective drug therapies for addiction were derived from the testing of compounds developed for other CNS diso...
متن کاملA Nurse with Pethidine Addiction
Pethidine is a synthetic opioid substance with high addiction potential. It is commonly used in medicine pre and postoperatively to relieve the pain. A Pethidine addiction generally starts as iatrogenic and may be a problem for the clinicians when there is a real pain. Its opioid analgesic usage is legal, yet it is possible that it can be abused by the health care workers and addiction may beco...

Background: Due to the lack of drug dependence prevention processes and the low effectiveness of treatment for the withdrawal of addiction and undesirable side effects of synthetic drugs, moving towards the use of reverse pharmacology to produce a less costly, effective, and low-risk drug is inevitable. Objective: Considering the long history of traditional medicine in Iran in the field of diag...

The present study was conducted to investigate the social status of addicts in Birjand City, Iran. This research was carried out by using the survey method. The research population included all adolescents and young people who were addicted to narcotics since the beginning or have turned to synthetic drugs before the age of 30. The samples were selected using the available sampling method. A re...

One area which has been largely neglected when studying the acquisition of addiction to smoking with the transtheoretical model is whether the individual had previously experimented with smoking. The importance of including the experimentation variable was supported by this research.
